Oh now I get it.
That was stupid yet funny. Those who don't understand the joke there are many people who sell stuff in New York. If they are selling T-shirts they would be yelling "T-shirts! T-shirts! T-shirts!" to attract attention. The guys was yelling "Puppets! Puppets! Puppets!" so it looked like he was trying to do business so that's why one guys asked him "How's business?" and when the guy replied "Business? What business?" The guy was basically making a fool of himself yelling out "Puppets!"
